15000 K562 cells with UPR sensor genes knocked out and treated with thapsigargin.

In UPR epistasis experiment, Perturb-seq was applied to explore the branches of the mammalian UPR. Using the three-guide Perturb-seq vector, sgRNAs targeting each UPR sensor gene were introduced into K562 cells with dCas9-KRAB. Transduced cells were then pooled, sorted for vector delivery, and after 5 days of total growth, treated with thapsigargin. Control cells were treated with DMSO. Transcriptomes of ∼15,000 cells were sequenced.
